EZCut™ TEV Protease for Affinity Tag Removal

With its stringent sequence specificity, TEV Protease is one of the best-characterized enzymes to chop off affinity tags from recombinant proteins. The currently available recombinant TEV proteases are riddled with disadvantages like autolysis, low activity and high costs.  BioVision is excited to release an enhanced version of this enzyme – the EZCut™ TEV Protease, which overcomes these issues and provides more stability than the native TEV protease, resulting in enhanced long-term activity.

LST: Lymphoid Screening Tube

The LST kit is the most complete 8-color screening tube for lymphocytosis studies. This 12-antibody combination has been designed by the EuroFlow™ consortium aiming at the detection of phenotypically aberrant populations of mature B-, T- and NK-cells.

- EuroFlow™ consortium validates the utility of LST combination to discriminate between normal and abnormal lymphoid cells with an overall efficiency of 96.7 %.

- Lymphocyte subset values belonging to a pathological sample. These are intended as representative values only. Each laboratory should establish its own expected values from the local population of healthy donors.

 

ALOT: Acute Leukemia Orientation Tube

The ALOT kit designed by the EuroFlow™ consortium contains a combination of 8 antibodies for initial assessment of the nature of immature populations of hematopoietic cells in samples with suspicion of acute leukemia. The ALOT kit also allows appropriate orientation towards a proper characterization panel (BCP-ALL, T-ALL and/or AML).

- An integrated analysis of the ALOT screening tube is proved to be an unprecedentedly strong tool for acute leukemia diagnosis. The ALOT combination shows an orientation efficiency of 98.3% for non-ambiguous lineage cases, which has been tested prospectively at different centers. (Van Dongen et al. Leukemia 2012; 26:1908–1975).

 

PCST: Plasma Cell Screening Tube

The PCST kit designed by the EuroFlow™ consortium contains a combination of 8 antibodies for identification, discrimination and enumeration of coexisting normal/reactive and aberrant plasma cell populations. The PCST kit is recommended for initial screening of Plasma Cell Dyscrasias.

  

* FIX&PERM® Kit contains 2 reagents: fixation medium (Reagent A) and permeabilization medium (Reagent B). Based on the extensive experience of the EuroFlow™ laboratories, the FIX&PERM® reagents were selected to fix and permeabilize cells for intracellular staining.

PetakaG3

PetakaG3 is an innovative bioreactor designed for mammalian cell culture in physiologic tissue conditions. The distinctive feature of PetakaG3 is that being a disposable device incorporates an automatic gas diffusion control system (AGD-CS) that works interactively with the cells growing and living inside, WITHOUT HYPOXIA CAMBERS, GLOVE BOXES OR THREE-GAS INCUBATORS

For cells cultured in PetakaG3 and incubated in regular 21% oxygen atmosphere the AGD-CS provides:

1) Media dissolved oxygen concentration within the limits of the physiologic oxygen tensions in physiologic tissues (from 75 mmHg down to 15 mmHg) monitorable with a dissolved oxygen fluorescent sensor dot. Because this advanced trait PetakaG3 is the safest, smallest, unique portable disposable hypoxia chamber, also available with integrated DO sensor.

2) Media dissolved CO2 concentration, enough to maintain a physiologic pH without external extra-supply ofcarbon dioxide gas.

3) Controlled media evaporation even at 2% to 10% atmospheric Relative Humidity (RH).

PetakaG3 is available in several options for culturing all kinds of cells from vertebrate and invertebrate organisms, especially suited for stem cells, primary cultures and tumor cells.PetakaG3 LOT & PetakaG3 HOT with hydrophilic surface developed for anchored dependent cells growth and PetakaG3 FLAT with a hydrophobic surface, not permissive of cell adhesion, for non adherent cells growth and 3D cellular clusters and aggregates.PetakaG3-Matrix is available carrying different surface molecular matrixes, such as Collagen, Vitronectin, Matrigel, etc. and also structural matrixes (nano-fibers mesh), or combinations of structural and molecular.

PetakaG3 allows cultures of two different cell types in the same device. Both cell layers are separated by 3mm of media, which is appropriate to study intercellular interactions through diffusible factors and exosome exchange. The PetakaG3™ FLAT bioreactor has all of the same size and structure characteristics as the PetakaG3™ HOT bioreactor, except that the interior surfaces are not treated to promote cell adherence to its walls. This special bioreactor is produced with a flat polymer with a very low surface tension. It is for suspended cell cultures such as; Blood cell preparation, Leukemia Cells, Hybridomas Cells, Special cells necessary for the synthesis of specific products for pharmaceutical applications.

PetakaG4 DO-Sense

The Petaka DO-Sense with O2 sensor Integrates all advantages of the Petaka LOT and Petaka-ET with the precision of the advance Dissolved Oxygen Monitoring technology of Ocean Optics, Inc.

PetakaG3 DO-Sense has a fluorescent Oxygen minisensor of approximately 4 mm in diameter built inside (RedEye® Oxygen Patches with the FOXY coating). The sensor has an insignificant thickness which does not affect cell growth and expansion.

This sensor allows either continuous or intermittent readings of the DO levels in the cell culture media across unlimited time span, in a non-intrusive manner.A microspectro fluorimeter and a specific friendly and intuitive software ( NeoFox® Phase Measurement System) emits light pulse flashes to the sensor and retrieves fluorescent responses from it. The processor of this system calculates the precise amount of DO through the intensity of the sensor emitted light and the exact temperature of the media. Read scientific foundation of the system. Bio-Synthesis, Inc., a leading biotech supplier of chemical biology reagents, is proud to announce Instant Quenchers™, a better alternative to BHQ 1, 2 and 3 for oligonucleotides.


Dual labeled oligonucleotides are commonly used to monitor the kinetics of PCR amplification and other biological processes in real time. In one version of this method, the oligonucleotides are designed to hybridize to the 3’ side (“downstream”) of an amplification primer so that the 5'-3' exonuclease activity of a polymerase digests the 5’ end of the probe, cleaving off one of the dyes. The fluorescence intensity of the sample increases and can be monitored as the probe is digested during the course of amplification.


In recent years TAMRA, as well as other fluorescent acceptor molecules in dual labeled probes, have been replaced with a growing family of dark quencher molecules such as our IQ™, Instant Quencher™ family of dyes. Quenchers are a class of chromophores which, instead of emitting absorbed fluorescence resonance energy as light, have the useful property of transforming the light energy into heat. This type of probe design can greatly simplify many fluorescence assays since there is no background fluorescence.


Fourth generation Instant Quenchers™, or IQ4™, have unique fluorescence quenching properties with an operational range between 500 nm and 700 nm and can therefore be utilized in multiplexed assays pairing IQ-4 quencher dye with different fluorophores. IQ4 can be used at either end, as well as an internal substitution during standard oligo synthesis. Description: PTEN, a tumor suppressor, has been implicated in a large number of human tumors and is conserved from humans to worms. Background
CD79a (Ig alpha, MB1) forms disulfide-linked heterodimer with CD79b (Ig beta). They both are transmembrane proteins with extended cytoplasmic domains containing immunoreceptor tyrosine activation motives (ITAMs), and together with cell surface immunoglobulin they constitute B-cell antigen-specific receptor (BCR). CD79a and b are the first components of BCR that are expressed developmentally. They appear on pro-B cells in association with the endoplasmic reticulum chaperone calnexin. Subsequently, in pre-B cells, CD79 heterodimer is associated with lambda5-VpreB surrogate immunoglobulin and later with antigen-specific surface immunoglobulins. At the plasma cell stage, CD79a is present as an intracellular component. CD79a/b complex interacts with Src-family tyrosine kinase Lyn, which phosphorylates its cytoplasmic ITAM motives to form docking sites for downstream signaling.

However, screening HDAC inhibitory compounds has proven to be difficult over the past due to the lack of convenient tools for analyzing HDAC activity. The new Colorimetric HDAC Activity Assay Kit provides a fast and convenient colorimetric method that eliminates radioactivity, extractions, or chromatography, as used in the traditional assays. The new method requires only two easy steps, both performed on the same microtiter plate. First, the HDAC colorimetric substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (e.g., HeLa nuclear extract or your own samples). Deacetylation of the substrate sensitizes the substrate, so that, in the second step, treatment with the Lysine Developer produces a chromophore. The chromophore can be easily analyzed using an ELISA plate reader or spectrophotometer. The assay is well suited for high throughput screening applications.
